BIOLOGY
CJC-1295 without DAC, also known as Modified GRF (1-29), is a synthetic analog of the first 29 amino acids of growth-hormone-releasing hormone (GHRH) — the shortest fully functional fragment of native GHRH. It contains amino-acid substitutions that improve resistance to enzymatic degradation while retaining activity at the GHRH receptor. Unlike the DAC (Drug Affinity Complex) version, the no-DAC form does not covalently bind albumin and therefore has a shorter duration of action.

CJC-1295 acts on pituitary somatotrophs via the GHRH receptor, triggering cAMP signaling that promotes growth-hormone synthesis and release. It has been studied in preclinical and clinical research models examining the somatotropic axis, pulsatile GH secretion, and downstream IGF-1 production, making it a valuable tool for in-vitro and in-vivo endocrinology research.

This product is intended solely for in-vitro and preclinical research use and is not for human or veterinary consumption.

KEY RESEARCH APPLICATIONS OF CJC-1295 NO DAC

1. GHRH Receptor Signaling
CJC-1295 No DAC demonstrates significant activity in endocrinology studies through:
• GHRH Receptor Activation: Binds and activates pituitary GHRH receptors.
• cAMP Signaling: Triggers intracellular cAMP cascades driving GH transcription.
• GH Synthesis & Release: Stimulates growth-hormone production and secretion.
Supporting Studies:
Teichman, S.L. et al. — Journal of Clinical Endocrinology & Metabolism. “Prolonged stimulation of growth hormone and insulin-like growth factor I secretion by CJC-1295, a long-acting analog of GH-releasing hormone.” PubMed – PMID: 16352683

2. Pulsatile GH Secretion & IGF-1 Axis
Investigated in somatotropic-axis research models through:
• Pulsatility: Studied for preservation of pulsatile GH secretion patterns.
• IGF-1 Downstream: GH stimulation studied for downstream IGF-1 elevation.
• Trough GH Levels: Examined for effects on inter-pulse GH concentrations.
Supporting Studies:
Ionescu, M. & Frohman, L.A. — Journal of Clinical Endocrinology & Metabolism. “Pulsatile secretion of growth hormone persists during continuous stimulation by CJC-1295.” PubMed – PMID: 17018654

3. GHRH Analog Pharmacology
Explored in broader endocrinology research through:
• Receptor Pharmacology: Reference GHRH analog for receptor characterization.
• Bioconjugate Comparison: Studied alongside DAC-modified long-acting analogs.
• Translational Endocrinology: Applied across pituitary and GH-axis research.
Supporting Studies:
Jetté, L. et al. — Endocrinology. “hGRF(1-29)-albumin bioconjugates activate the GRF receptor on the anterior pituitary in rats: identification of CJC-1295 as a long-lasting GRF analog.” PubMed – PMID: 15817669

RECOMMENDED RESEARCH CONCENTRATIONS
For laboratory research reference only. Not for human or veterinary use.
• In-vitro pituitary cell assays — 1-100 nM — Assay buffer
• Receptor signaling studies — 1-50 nM — Cell culture media
• Animal models (rodents, SC) — Per protocol — Injection
